چکیده

Blast disease resulting from Magnaporthe oryzae fungal infection reduces annual rice yield by up to 30% globally. The wheatwin2 (wwin2) is a pathogenesis-related (PR) gene that encodes for PR-4 protein with chitinase properties capable of degrading chitin, major constituent certain cell walls. However, the potential wwin2 contribute M. resistance in unclear. This study reports construction pMDC140 vector carrying and its Agrobacterium-mediated transformation into Tadong genome. In brief, was synthesized integrated using Gateway cloning technology transformed Our results show promising high rate, more than 90% calli expressing β-glucuronidase (GUS), reporter marker. expression further confirmed quantitative real-time polymerase chain reaction. conclusion, pMDC140-wwin2 constructed, which had rate could consistently induce GUS genes rice. Data this beneficial subsequent vitro oryzae-infected field experiments confirm defense mechanism towards blast

A Preliminary Experiment on Agrobacterium tumefaciens-Mediated Transformation of the P5CS1 GENE in Tall Fescue

Abiotic stress conditions can be imposed on plants through either poor water quality or adverse climate condition. These types of stress affect plant growth and development throughout their life cycle. Plants that are tolerant to drought are therefore valuable, and turf grasses have the potential to grow tolerant. Turf grasses provide ground cover and offer multiple benefits, from their aesthet...

Agrobacterium-mediated transformation of cotyledonary leaf of lettuce (Lactuca sativa L.) by the GCHI gene

Lactuca sativa is widely used as a leafy vegetable belonging to the Asteraceae family. Cotyledonary leaves of in vitro lettuce seedlings were transformed by plasmids containing the GCHI gene. Also the effects of number of days of pre-cultivation and co-cultivation of explants were examined on the transformation frequency. After co-cultivation, the explants were transferred onto the regeneration...

Agrobacterium-Mediated Transformation of Pomegranate (Punica granatum L.) ‘Yousef Khani’ Using the gus Reporter Gene

In this study, an efficient Agrobacterium-mediated transformation method was developed forpomegranate (Punica granatum L.), a difficult-to-transform plant. In vitro shoot segments wereinoculated with Agrobacterium tumefaciens strain LBA4404 harboring the binary vectorpBI121 carrying the neomycin phosphotransferase (nptII) gene as a selectable marker and β-glucuronidase (gus) gene as a reporter....

Optimization of Agrobacterium-mediated transformation in oyster mushroom (Pleurotus ostreatus) by vector containing human pro-insulin gene

Transferring foreign genes into mushroom mediated by Agrobacterium tumefaciens is a standard technique in genetic engineering. Recombinant human insulin has been greatly used in the treatment of type I diabetes. The production of edible mushroom derived insulin should facilitate oral delivery. In this study we used the Agrobacterium tumefaciens mediated transformation method for the transfer an...
